Description:
EXPECTED COMPETENCES TO BE ACQUIRED:
1. To distinguish personal, systematic and random errors.
2. To calculate basic statistical quantities (mean, median, standard deviation).
3. To explain the principles of gravimetric analysis.
4. To explain the dissociation of acids and bases as well as hydrolysis and solubility of salts in aqueous solutions using corresponding equilibrium constants.
5. To calculate pH of the aqueous solution of an appropriate chemical composition.
6. To distinguish the equivalence point and the titration endpoint. To define the primary and secondary standard.
7. To calculate the amount of an analyte based on the data of gravimetric and volumetric measurements.
8. To explain the principles of potentiometric measurements.
9. To explain Beer's law and calculate the amount of an analyte based on the spectrometric measurements.
10. To explain chromatographic methods for analysing complex samples and interpret chromatogram.

COURSE CONTENT:
LECTURES
1. The role of analytical chemistry, introduction to chemical analysis from sampling to analytical information.
2. Errors in chemical analysis. Statistical data treatment and evaluation.
3. Gravimetric methods (precipitate and reagent characteristics, application of gravimetric methods).
4. Chemical composition of water solutions and chemical equilibrium.
5. Titrimetric methods (volumetric titrimetry, standard solutions and calculations in volumetry).
6. Theory of acid-base titrations (solution and indicators, neutralization curves for strong acids and bases, neutralization curves for weak acids and bases, buffer solutions).
7. Precipitation titrimetry with silver nitrate and complexometric titrations.
8. Fundamental principles and application of redox titrations.
9. Potentiometric methods (reference and indicator electrodes, direct potentiometry, potentiometric titrations).
10. Introduction to spectrometric methods and instruments for optical spectrometry.
11. Molecular absorption spectrometry (UV-Vis, IR).
12. Molecular fluorescence spectroscopy.
13. Atomic spectroscopy (absorption and emission).
14. Introduction to chromatographic methods. Gas chromatography (principles and applications).
15. Liquid chromatography (principles and applications).

LABORATORY EXERCISES
1. Determination of cations.
2. Determination of anions.
3. Titrimetric determination of chlorides by standard solution of AgNO3 (Fajans method);
Titrimetric determination of bromides by standard solution of AgNO3 (Volhard method).
4. Determination of calcium and magnesium in mixture by complexometric titrations with EDTA.
5. Spectrophotometric determination of KMnO4.
6. Determination of acetylsalicylic acid in pharmaceutical products by IR spectroscopy.
7. Spectrofluorimetric determination of quinine.
8. Determination of sodium by flame photometry.

3. Chemical equilibrium and calculation of the concentration of solutions.
4. Calculation of solubilities.
5. Buffer solutions. Calculation of the pH values.
6. Calculation of results from volumetric data.
7. Titration curves (neutralization, complexometric, redox and precipitation titrimetry).
8. Beer's law.
9. Calculation of results from spectrometric data.
10. Calculation of chromatographic quantities (resolution, selectivity factor, etc.).
Literature:

Klasične (gravimetrijske i titrimetrijske) metode analize; instrumentne metode analize: elektrokemijske metode (potenciometrijske i nepotenciometrijske metode); spektrometrijske metode (IR, UV-Vis, fluorimetrija); kromatografija (poseban osvrt na kromatografiju biomolekula) te imunoanalitičke metode (precipitacija u gelu, aglutiniranje, fluoroimunoanaliza, enzimska analiza). Vježbe: kvalitativna analiza biološki važnih iona; kvantitativna analiza (titrimetrijske metode - priprava standardnih otopina i analiza uzoraka različitog sastava); IR i UV-Vis spektrofotometrija; spektrofluorimetrija; plamena fotometrija; imunoprecipitacija u gelu (određivanje imunoglobulina u humanom serumu).
