Course objective:
Explain the advantages and disadvantages of using high-throughput sequencing technologies with an emphasis on recommendations when designing experiments, with the aim of obtaining quality data for downstream analysis
Content:
Overview of the most commonly used high-throughput sequencing technologies (whole-genome DNA sequencing, RNA sequencing, genome methylation analysis, characterization of DNA-protein interactions, metagenomics and metatranscriptomics, single-cell mRNA and DNA sequencing)
- Recommendations for designing genome-wide DNA sequencing experiments and targeted resequencing of parts of DNA
- RNA sequencing: recommendations in the design of transcriptome, microRNA, long non-coding RNA and ribosome profiling experiments
- Recommendations when designing an experiment in which methylation profiles are determined, design of a ChIP-Seq experiment
- Design of metagenomic and metatranscriptomic experiments
- Design of single-cell mRNA and DNA experiments
- Designing HiC experiments
